Design Notes
Each of the parts were cleaved with Endonuclease and ligated with pSB1C3 vector. First, a promoter and a reporter gene were amplified by PCR and two of the pairs were ligated with pSB1C3 vector. Second, another promoter and reporter gene were put together by PCR and then ligated with pSB1C3 vector with the other two-pair-fragment. Three pairs were finally put in a vector and it underwent point mutation during the procedure because it had multiple cleavage sites.
